Ultra-sensitive immunoassay technologies

Two high-quality automated immunoassay platforms are available in the Karikari lab: four Simoa HD-X™ analyzers (Quanterix Corp.) and a Lumipulse G1200 (Fujirebio Inc.). The HD-X analyzer is an ultrasensitive immunoassay platform that measures minute levels of target analytes in diverse sample matrices, including blood and cerebrospinal fluid (CSF).. Our extensive experience in this ultrasensitive immunoassay platform has led to practical proficiencies in developing our in-house blood assays from scratch. Notable blood assays developed together with our collaborators at the University of Gothenburg include brain-derived tau, p-tau181, p-tau217, p-tau231, neurofilament light, total tau, glial fibrillary acidic protein, and neurogranin, some of which are now commercialized and widely used worldwide. The Lumipulse G1200 is a high-throughput and fully automated chemiluminescent enzyme immunoassay system that utilizes unique mono-test immunoreaction cartridges and can run up to 120 tests/hour. Several Lumipulse assays have been validated for routine clinical measurement of the core AD biomarkers, including Aβ42/Aβ40, p-tau181, and total-tau.
